| PRINCIPLE
Aklides is a multicomponent
system:
- a high quality fluorescence microscope
- a LED lightsource (autonomy > 100000 hours)
- a motorized scanning stage
- a CCD camera
- Aklides® software
The Aklides®
software is the heart of the system, providing the algorithms
for optimal image generation (positioning, focusing, quality
control), interpretation and pattern recognition. Fluorescence
intensity is quantified in arbitrary units, allowing better
standardization in autoimmunity testing. Images and results
are saved in the system data bank and included into patient's
reports at the pdf format. Results can be easily exchanged
between laboratories.
Image acquisition in the
standard mode takes less than 1 minute per well for HEP-2
testing, including pattern recognition. A screening mode for
differentiation between positive and negative samples allows
processing of 64 wells within a few minutes.
In its current version,
Aklydes has 4 different modules:
- ANA (HEp-2): positive/negative interpretation and pattern
recognition.
- nDNA (Crithidia lucilliae): positive/negative interpretation.
- ANCA: differenciation between c-ANCA and p-ANCA.
The software is user-friendly:
the user chooses the slide format, key in or import patient
data, loads the slides (up to 4 in the standard configuration)
and starts the reading. The complete process takes between 15
minutes (nDNA) and 60 minutes (HEp-2) for 4 slides. Each
result has to be validated (or modified) by the user before
storage in the data bank.
All pictures and results are
automatically saved and can be extracted to generate several
reports (per slide, per patient, per autoantibody, etc).
Teaching hospitals will also greatly benefit from the system.
Aklides® software has been
developped for reagents specially optimalized for the system (list
on the right), although it can be used as a conventional
microscope in open mode, but without automated slide
processing. In this configuration, the user can take the
pictures of his choice without the need of a dark room and
store them in the system archives. |
